Abstract:

Background Although spermatozoa delivers its RNA to oocytes at fertilization, its biological role is not well characterized. Our purpose was to identify the genes differentially and exclusively expressed in sperm samples both before and after the swim-up process in control donors and infertile males with the purpose to identify their functional significance in male fertility. MaterialsAndMethods This was a nested case-control study. Ten sperm samples were obtained from infertile patients [n=5 (two aliquots each from five samples; one before the swim-up process and one after)] and donors [n=5 (two aliquots from five samples, one before the swim-up process and one after)]. Oligonucleotide microarrays were employed to study the genome-wide expression of pooled samples from infertile patients vs. donors. A total of four microarrays were perfomed: two with sperm sample aliquots before swim-up and two with sperm samples aliquots after swim-up, from both the case and control groups. The results were evaluated to detect which genes expressed differentially [fold change (FC)>5 and p

Sperm retrieval techniques for assisted reproduction.

Different surgical methods such as PESA, MESA, TESA, TESE and micro-TESE have been developed to retrieve spermatozoa from either the epididymis or the testis according to the type of azoospermia, i.e., obstructive or non-obstructive. Laboratory techniques are used to remove contaminants, cellular debris, and red blood cells following collection of the epididymal fluid or testicular tissue. Surg...

Sperm Preparation Techniques for Artificial Insemination - Comparison of Sperm Washing, Swim Up, and Density Gradient Centrifugation Methods

The Artificial Insemination (AI) is the first option treatment for infertile couples with cervical factor subfertility, mild-moderate male subfertility and unexplained infertility. With the exception of cases in which the use of in vitro fertilization (IVF or ICSI) is strictly due as a consequence of a severe male or female factor, the artificial insemination must be part of a gradual approach ...

Comparative analysis of three sperm DNA damage assays and sperm nuclear protein content in couples undergoing assisted reproduction treatment.

STUDY QUESTION Is there an association between sperm DNA damage, measured by three different assays, sperm nuclear protein content and clinical outcomes in assisted reproduction treatment (ART)? SUMMARY ANSWER Sperm DNA damage measured by terminal deoxynucleotidyltransferase-mediated dUTP nick-end labelling (TUNEL) and the Comet assay were significantly associated with ART outcomes in our sin...

Ficoll-400 density gradient method as an effective sperm preparation technique for assisted reproductive techniques

BACKGROUND Infertility being a burning issue, the male itself contributes about 40% as a cause, as evident by statistical data. However, Assisted Reproductive Technology (ART) has emerged as a powerful tool in the management of infertility. Sperm preparation techniques govern the selection procedure to separate functional spermatozoa which can then be used in IUI, IVF, and ART and for cryoprese...

New Trends in Assisted Reproduction Techniques: Cryopreservation, In vitro Fertilization, Intracytoplasmic Sperm Injection and Physiological Intracytoplasmic Sperm Injection

Since that historically the first human birth was registered through in vitro fertilization in 1978, the improvement and creation of new assisted reproduction techniques such as intracytoplasmic sperm injection and physiological intracytoplasmic sperm injection have become of great importance nowadays. Louise Brown was the first baby conceived through in vitro fertilization in England. Since th...
